Biosynthesis of dihydroxyardeemin by heterologous expression

Ardeemins are a group of indole terpenoids from fungi and important bioactive natural products. The ardeemin biosynthetic genes ardudgABCD from Aspergillus udagawae were expressed in Aspergillus oryzae, to produce ardeemin FQ (1), dia-ent-ardeemin FQ (1a), 5-N-acetylardeemin (3), 5-N-acetyl-16ahydroxyardeemin (4), 5-N-acetyl-15b-0-hydroxyardeemin (4a), and 5-N-acetyl-15b-0,16a-dihydroxyardeemin (5). In the course of our studies of novel catalysts, we identified a new a-ketoglutarate oxygenase, ArdD, which hydroxylates two different positions of the pyrroloindoline skeleton.
