Imaging mass spectrometry (MS) aims to correlate localization of molecules in with sample morphology. In order to do such correlation, a thin tissue section is mounted on a target plate similarly to a histological sections. Tissue sectioning is the first steps for most of the imaging MS experiment and should be optimally performed in order to avoid delocalization or degradation of molecules within the tissue. Tissue sectioning requirements and strategies for imaging MS are discussed in this chapter.
