•ATP-dependent Lon proteases are key components of the protein quality control systems.•More than seventy structures of different Lons and their fragments have been determined.•This structure-based review summarizes the current state of knowledge of Lon proteases.•Lons are potential drug targets that require understanding of structure and function.
