Trace Mirna Assay Based On Dna Nanostructures Formed By Hybridization Chain Reaction And Gold-Nanoparticle Tags

microRNAs (miRNAs), a class of endogenous non-coding RNAs, are involved in many crucial biological processes, which have emerged as a new set of biomarkers for disease theranostics. Exploring efficient signal amplification strategy is highly desired to pursue an ultrasensitive miRNA biosensing platform. In this work, we have developed a novel electrochemical approach for the analysis of trace miRNA. Target triggered DNA assembly is performed on the surface of the electrode. Meanwhile, gold-nanoparticle tags are integrated on the product of hybridization chain reaction, which further enhance electrochemical response towards initiator miRNA. The as-prepared electrochemical biosensor shows good analytical performances and satisfactory practical utility. It has great potential to provide a powerful tool for the miRNA assay in biomedical fields.
