•CII and aspirin acted synergistically to disrupt the intestinal mucosal barrier.•Aspirin and CII shaped innate and adaptive immune reactions into Th1-dominant.•The damage of mucosa barrier is prerequisite for CII-induced intestinal inflammation.•This study provides a new strategy for treatment and prevention of IBD.
