Longitudinal Cortical Thickness Analysis in Alzheimer Disease. ADNI-Arg Cohort (P3.188)

Objective: The aim is to assay longitudinally the cortical regional changes in CT in 3 different groups: healthy controls (HC), Mild Cognitive Impairments (MCI) and Mild AD. Background: Cortical Thickness (CT) is a new method that allows measuring cortical changes in Magnetic Resonance Images (MRI). Although, there have been previous publications related Alzheimers’ Disease (AD), this is the first Latin-American cohort that was studied in a uniform manner and defined clinically and biologically with AD biomarkers. Design/Methods: Fifty-six participants were selected. All of them have undergone clinical and cognitive assessments, brain MRI scan and AD biomarkers during the initial visit. Participants were separated into the three clinical groups: 15 HC, 27 MCI, and 14 AD. MRI scans were repeated at month 6, 12 and 30 during follow-up. Differences in CT were analyzed between groups and the variability during follow-up. Results: Significant differences were seen between AD and MCI versus HC in the medial and inferior temporal lobe, parietal, precuneus and occipital regions at both hemispheres at the cross-sectional analysis. In the longitudinal assessment, hippocampi have shown less variability than adjacent regions (para-hippocampus), especially in those with positive AD biomarkers. Conclusions: Cortical Thickness might be used to differentiate clinical groups related with AD and we identified more useful measurements not only at the hippocampi but in adjacent regions.
