•C. reinhardtii accumulated similar levels of Cr(III) and Cr(VI).•The subcellular partitioning of Cr(III) and Cr(VI) was similar.•Cr(III) and Cr(VI) associated mainly with organelles and heat-stable proteins.•Metallomic analysis showed two main Cr-binding biomolecules after 72h of exposure.
