•Serum HMGB-1 is positively associated with proinflammatory cytokine levels in PRRS.•Upregulation of HMGB-1 and proinflammatory cytokines mRNA in lungs of PRRSV infected piglets.•HMGB-1 plays an implicit role in PRRS pathogenesis.•HMGB-1 may constitute a potential therapeutic target for treatment of PRRS in pigs.
