•The pickup and delivery problem with transshipment is studied.•A large neighbourhood search and a genetic algorithm are proposed for the problem.•A tree-shaped buffer based cache memory is defined to accelerate the decoding phase.•Both algorithms show better results than in the literature.•The genetic algorithm outperforms the large neighbourhood search.
