•Mutational analyses of the Tvp38/DedA family member YqjA show that inactivation does not affect oligomerization.•YqjA forms dimers in the membrane and the C-terminal transmembrane region is involved in homo-interaction.•The inactive variant YqjAE39A/D51A is still capable of dimer formation.
