[Osteogenic growth peptide regulates proliferation and differentiation of calvarial osteoblast-like cells of rat].

OBJECTIVE:To investigate the effect of osteogenic growth peptide(OGP) to the proliferation and differentiation of osteoblast-like cells in vitro. METHODS:Calvarial osteoblast-like cells of rat bones were cultured individually in medium contained various concentrations (10(-11) mol/L-10(-7) mol/L) of OGP; and the cell proliferation was checked by MTT method, alkaline phosphatase activity was detected by ELISA and level of Cbfal mRNA was amplified with RT-PCR. RESULTS:In vitro, the OGP put in culture medium resulted in an increase of cell proliferation (P < 0.05), ALP activity (P < 0.05) and level of mRNA for Cbfal (P < 0.05) respectively. All used OGP concentrations (10(-11) mol/L-10(-7) mol/L) were effective but the maximum efficiency was obtained with 10(-8) mol/L OGP. CONCLUSION:Our results demonstrate that OGP stimulates proliferation and sequential differentiation in vitro of calvarial osteoblast-like cells of rat bones.
